IGF signaling is associated with decreased cell death and increased cell proliferation. Activation of IGF-1R leads to the activation of two pathways, PI3K/AKT and Ras/MAPK. PI3K/AKT pathway ultimately leads to decreased apoptosis, increased glucose metabolism, increased protein synthesis. On the other hand Ras/MAPK pathway leads to increased cellular proliferation by activating different transcription factors, such as ELK1. Cancer cell proliferation, migration and invasion have been related to the activation of IGF-1R in many studies previously (Sachdev and Yee 2007). Cancers in lung, breast, colon, prostate are showed to be associated with increased level of plasma IGF (Pollak, Schernhammer and Hankinson 2004). Other members of IGF pathway are also proved to be involved with malignancies. This provides a way to treat cancers via modulating IGF pathway and various ligands involved with it. Drug resistance by tumors is one of the critical conditions in new age cancer treatment. As IGF pathway is involved in cell proliferation it shows a new era of treating these kinds of situations. Many studies have shown that modulating IGF pathway has promising effect in these cases (Denduluri et al. 2015). Ovarian cancer is the fifth most frequent cause of cancer death in women. In nearly half of ovarian and breast cancer patients, RAB25 mRNAs were found to be selectivey overexpressed at stage 3 and 4 serous epitheli al ovarian cancers in comparison to other genes located in the amplified region. Decreased expression of two proteins BAK and BAX were found to be associated with an inhibition of apoptosis in such cells. Simultaneously activation of PI3K/AKT pathway provided potential mechanisms to employ the effects of RAB25 on tumor aggressiveness (Lee et al. 2015). Involvement of PI3k/AKT pathway in the RAB25 related tumor aggressiveness leads to the way to treatment by modulating this pathway. Recently various evidences show that IGF-1 promotes atherosclerotic plaque stability and consequently reduces atherosclerosis in animal models. IGF-1 induces reduction of oxidative stress, cell apoptosis, proinflammatory signaling, and endothelial dysfunction, that helps prevent the disease. Aging has been widely linked with increased vascular oxidative stress and certain vascular diseases (Barzilai et al. 2012). It has been postulated IGF-1 may play a crucial role on vascular aging processes. There are various evidences that IGF-1 exerts pleiotropic effects on the vasculature resulting in reduced vascular oxidative stress, apoptosis, and inflammatory signaling pathway (Higashi et al. 2012). By modulating IGF signaling disease related to stress or premature cell death or decreased cell death can be prevented in future. Further studies on signaling pathway of IGF show its potential role in regulation of autoimmune diseases. Though this area of treatment requires a lot more research and practical approaches, the role of IGF signaling is competent enough. Studies show that IGF-I can potentially protect animal models from developing insulin-deficient diabetes mellitus (Gao et al. 2011). On the contrary, patients with Graves disease have shown an overexpression of the IGF-1R in many cell types and antibodies against the receptor were prevalent in such patients. The frequency of IGF-IR positive B and T cells were found to be relatively high in patients with the disease. Hence, as IGF-1 and IGF-1R is found to be linked with the mentioned autoimmune disease modulation of the pathway might be a potential therapeutic target for managing the diseases and alleviating the health consequences (Smith 2010). The modulation of IGF pathway opens new ways to treat various diseases like different types of cancer, cellular stress-related diseases and immunological diseases also. It depends totally on massive research and practically showing the results on animal models to prove the modulation of IGF pathway actually showing some positive results. Comparative Genomic Hybridization: Comparative Genomic hybridization is a comparative study of two genomes by in situ hybridization technique that allows detection of differences in DNA sequences between the two genomes. Two genomes, one study and one reference genomes are differentially labeled after being denatured into single stranded DNA (Pinkel and Albertson 2005). These two genomes are then hybridized to normal metaphase spreads or to microarray. Ratio of the fluorescence intensity are then measured And plotted on a graphical manner. The graph can reveal locations of copy number changes in along the target chromosome or DNA sequences. It has been extensively used to identify genetic differences between normal cells and certain diseased cells mostly in cancer cell biology. It efficiently detects any gain or loss of genetic material in human genetic disorders by comparing the same with a reference genome, often a relevant normal cell genome. Primarily the effect of stable expression of Rab25, a hallmark of cancer has been assayed in all the parts of Figure 2. Other hallmarks that were also assayed were Bak and Bax nuclear proteins. Bak and Bax are two nuclear-encoded proteins that are capable of moving through mitochondrial membrane and induce cell death by apoptosis. Phosphorylation of AKT was also assayed during stable expression of Rab25 and in presence of RNAi. AKT or protein kinase B (PKB) is a protein kinase that plays key roles several cellular functions including apoptosis. Viral-mediated transduction is the most potent technique to produce stable cell lines with a overexpressing protein. Vectors are utilized for this purpose. Vectors are autonomously replicating DNA molecules that can be used to transfer DNA fragments to cells of interest. For transfecting mammalian cells, vectors derived from mammalian viruses are used that include Simian viruses (SV40), polyomavirus, herpesvirus and papovirus (Khan 2013). The Rab25 gene can be inserted into any one of the vector mentioned and the same gene can be transferred to an appropriate mammalian cell line by viral transfection. Mammalian cell lines that are extensively used for this purpose are HeLa, HEK293T, U2OS so on and so forth. Compared to transient transfection, stable transfected cell lines are produced to analyze impact of altered gene expression over a long time period. Generally, antibiotic resistance or fluorescent reporter cell markers are incorporated into plasmid or vector DNA to create selectio n criteria. Clonal Selection can be done by appropriate selection controls. The selected colony is used to produce monoclonal cell line development and consequently producing a stable cell line generation. Bak protein levels are found to be lower in Rab25 expressing A2780, HEY and T29 cells as compared to pcDNA cells. In T80 cell lines Bak was overexpressed showing that Bak levels are regulated by other factors in certain cells. Bax levels were low in A2780 and Hey cells but not in T29 and T80 cells. In assay (f) Bak and Bax levels were measured in A2780 cells were Rab25 expression is downregulated by RNAi. The downregulation of Rab25 nullified Rab25 mediated inhibition of Bak and Bax and hence the two protein levels were found to be high in the cell lines. Bak and Bax are two nuclear proteins that can travel to mitochondrial outer membrane upon activation of relevant signaling pathway and is essential for mitochondrial permiabilisation during apoptosis (Dewson and Kluck 2009). These two proteins play a significant role in intrinsic apoptotic pathway in mammalian cells and as a result they have being extensively studied for therapeutic purposes to block apoptosis. Motility of cells where Rab25 is overexpressed or knocked down can be assayed by standard apoptotic assay methods. Cleavage of the genomic DNA into discrete fragments and consequently membrane disintegration are key features that occur during apoptosis and cell-mediated cytotoxicity. DNA cleavage being hallmark of apoptosis is often measured to assay apoptotic cell death. The DNA fragments can be assayed by ELISA quantification of histone-complexed DNA fragments. Furthermore, caspases are a class of protein that are expressed and involved in the early stages of apoptosis. Activation of the caspase proteins can be assay to determine the stage of apoptosis in a cell (Suzuki, Nakabayashi and Takahashi 2001). This can be done by in vitro enzyme assay or by detection of cleavage of an in vivo caspase substrate. Furthermore to assess the motility transwell migration assay can be performed to assess cell movement and migration. In Transwell migration assay there are two chambers; the cell s uspension placed on the upper chamber and chemoattractants are placed in the lower chamber. At the end of Act 3, scene 5, just after the big argument between Capulet and Juliet, the Nurse is trying to comfort a distraught Juliet, but instead she fails her miserably. When Juliet asks the Nurse what she thinks she should do, the Nurse surprises her by saying that she might as well marry Paris because Romeo has been banished and they can no longer be together. She thinks that love (marriage) is no more than having sex and having babies, then to her Paris would make just a good husband as Romeo would. She does not understand the depth of emotion involved in true love, and Juliet is very upset by this because she thought that the Nurse understood how she felt. After this conversation, Juliet goes to the Friar for advice (Act 4, scene 1). The Friar helps her a great deal, and you think he really understands Romeo and Juliet, but he in turn fails Juliet in Act 5, scene 3. When Juliet wakes up and find Romeo dead she wants to spend more time with him, but the Friar insists that they leave when he hear people coming their way, Come Ill dispose of thee among a sisterhood of holy nuns. The Friars inadequacy in this crisis is shown by this ironic suggestion. He is fine until now to help the lovers be together, but when it comes to him nearly being caught out, he just tries to run away from the situation. In this play of overflowing love and passion we are introduced to a character very different from all the others Paris. Paris is an altogether good man, who is genuinely in love with Juliet and is the only one genuinely upset when she dies. Here there is another situation of unrequited love for Juliet from Paris. He is quite an innocent character caught up in the Capulet familys complications and does not think there is anything wrong with the arranged marriage. He does not actually speak to Juliet until they meet in the Friars cell in Act 4, scene 1, and the marriage is arranged through Capulet. He is very thoughtful towards Juliets feelings, and in lines 6 and 7, he says he has not talked to her about love (the marriage), because she has been weeping and is very sad about Tybalts death, but thinks that by getting married she will be happy again. Another character apart from Romeo and Juliet who is also driven by great passions and love is Tybalt. Tybalt shows the greatest passion for hate. In the balcony scene (Act 2, scene 2), you can tell that they both see their love as more important than any ancient family feud. In lines 66-69, Romeo speaks of how stony limits cannot hold love out, meaning that even high walls are no challenge for love and that nothing can stop them from sharing their love. By the end of the play Romeo and Juliet have changed quite a bit. Romeo went from being almost in a state of depression, to this fiery most positive character. Urrea goes into great detail to describe each member so you feel closer to them and you are able to better understand their decisions. All of the men that chose to go wanted a better life, whether it is for themselves or for a loved one. These men all acted selflessly. If I were in their situation I would try harder to look for a job in Mexico, but if I could not find one, I suppose that I would do what they did. It costs the United States and Mexican government a lot of money to deal with these illegal entrants. However, neither one of them really do anything to try to stop it. I think that this was an isolated incident, but there could be more to come in the future. If I were the leader of Mexico, I would vow to fix the state of the country and make it so that people don’t have to immigrate to other countries to find work. If I were the president, I would want to tighten down on Border Patrol so this doesn’t happen again. I like the tone the author uses. I think it fits the message that the author is trying to relay to the audience about the story. The style was alright, it was a little hard to read. There were too many details packed into single sentences. The style made the book hard to read. In my personal opinion, there were too many facts, but I can see why they were necessary. I think the simple fact that these men migrated to make theirs and their family’s lives better is what impacted me the most. It really changed my view on illegal immigration. The said six major scopes of the said assessment device are the problem solving, communication, roles, effective responsiveness, effective involvement and behavioral control. The dimension on Problem Solving intends to measure the capability of a family to be able to face and solve certain problems that arise within the family. With the use of questionnaires having four possible responses on agreeing to the solutions suggested and disagreeing, each family is scaled depending on their answers. The other dimensions as well follow the same pattern of questionnaires. After answering the questions, each result shall be averaged, evaluated and concluded regarding the improvements of the family in the therapies they undergo. The standard results for the Family Assessment Device which concludes that the family has already improved through the family therapy is based on how well they have comprehended with the activities they undergo during the therapy. There are major ways on how to evaluate the changes in a family. One is through the statistical significance of changes observed, another is through the percentage of dropping below clinical cut-offs and finally through the magnitude of change between intake and termination. These three major evaluation processes helps the psychologists to scale the improvements that the family being observed has already incurred. Many experts have proved the reliability of tests and family assessments such as the Family Assessment Device effective. True, at times, the application of this assessment may not be that easy but the cooperation of the family under therapy programs would really help in making assessments such as this a successful program. At times, there might be some family members who would rather depend on their own understanding of the problems they are facing than referring their problems to experts. Usually, this makes the FAD application harder to apply, but as said earlier, constant assistance and assurance to the families that they are being helped and not exploited or whatsoever, the Family Assessment could be concluded to be successful. Conclusion As it could be noted through the studies and discussion presented above, the result of violence and abuse upon the victim simply depends on his or her perception of the situation that occurred. Someone who is strong and wise enough in facing life’s challenges may look at this particular experience as a lesson that would teach him to take full responsibility of the situation the next time that the same thing happens. However, a person who is actually frail in facing the surprises that life has to offer would break down upon experiencing certain devastating incidents. Yes, it is one’s development as a person and perception towards life and the challenges that it brings that would actually determine the ways by which an individual would be affected by violence or abuse. It is only through a person’s discretion that the effects of the said situations could be identified within the growth progress that that particular person would undergo. It is also upon the person’s discretion as to how he or she would actually allow the said incident to impact her personality as well as her social dealings.
